Biography
Jo surprises Dean by coming up behind him with a rifle in his back in Everybody Loves a Clown. He knocks her gun from her hands, but she punches him to reclaim it. Ellen shows up from the kitchen holding Sam at gunpoint. When Ellen realizes the boys were Dean and Sam Winchester, she drops get handgun so Jo drops her rifle as well. After introductions, Ellen talks with the boys while Jo listens in on the side. They point the boys over to Ash to help track the demon. Jo assures them he's a genius. After Ash leaves to go work his magic with the computer, Jo and Dean end up off to the side. He asks about her mom getting in the lifestyle, and she tells him about her dad being a hunter and passing away. Dean starts to flirt with her, but backs off. Jo's impressed. Normally cheap pickup lines, augmented with food, beer, and music and they think they can get in her pants. Dean leaves to go on a job. After the boys come back, Jo and Dean wait while Sam gets the hint to leave. Jo starts flirting, but Dean is honest with her and turns her down gently. Ash comes out with his setup, and promises to call the boys when he catches something. Dean and Sam leave.
Dean Smith has a little sister Jo in It's a Terrible Life. Zachariah erased Dean Winchester's memories, and gave him new ones, in which he lived a normal life.
In Heaven, the boys tell Ash that Ellen and Jo died fighting the good fight in Dark Side of the Moon.
In the alternate timeline when the Titanic doesn't sink, in My Heart Will Go On, Jo is alive and hunting something big with her own crew out in California. Massive amounts of people are dying, and they can't do anything to stop it. When the timeline was put back, Jo is again dead.
Appearance
Long blonde hair and petite, Jo is strong. She has brown eyes.
Personality
Spunky, full of fire and spit, Jo is also tenacious when she wants something and will not let it go. She had self confidence and self respect, not wanting anyone to walk over her, including her mom. She's emotionally aware to back off from Dean when he was hurting, and to help her mom come to the realization that Jo wasn't going to make it.
Equipment
She owns a knife, with her dad's initials engraved on it.
Powers and Abilities
Human Abilities
She can shoot, throw a knife, and fight. She's smart, and can put together a case and solve it. She's also a great barmaid.
Spirit Abilities
Her spirit was forced to try to kill Dean, even though she didn't want to. She was able to opn a window, turn on the gas in the gas stove, and grab Dean's lighter.
Weaknesses
She falls for Dean, but doesn't want anyone to know.
Deaths
Technically, she died once and was erased from existence once. When she died after being mauled by a hellhound, she was held by her mother. Ellen let the hellhounds into the store and, once she knew they were in the store with her, she hit the button on the bomb and blew them all up.
She was brought back in an alternate reality when the Titanic didn't sink. However, when that was fixed, she was erased from existence.

- In early drafts of the script for Everybody Loves a Clown, Jo was called "Alex".
- The writers spent so much time with a father-son relationship dynamic, that they wanted to explore a mother-daughter dynamic, which is where Ellen and Jo came about. - Supernatural Companion Season 2, pg 14

We wrote her as an innocent girl who wanted to be a hunter, and that gave her this energy of leaping before she looked and doing all these things that Dean wouldn't necessarily spark to. - There was a Jo's Blog on the Hunter's Blog official website from Warner Brothers, which redirected you to CW Supernatural page. It ceased being updated in early May 2007, after 2.18 Hollywood Babylon aired. It has since been removed from the websites. There were diary entries from Ho detailing her desire to become a hunter, and a few pictures from her Hunter's Blog. She quit it June 16, 2006, frustrated with herself because there's nothing new to say. She hopes to go on a solo hunt, and continue it after that.
- According to the original version of Jo's Blog, she knew John Winchester as early as 1994 and called him "Uncle John".
